  • Refereed, Molecules, MDPI AG, Cd2+-Selective Fluorescence Enhancement of Bisquinoline Derivatives with 2-Aminoethanol Skeleton, Yuji Mikata; Aya Tsuruta; Hinata Koike; Sunao Shoji; Hideo Konno, The development of fluorescent Cd2+ sensors requires strict selectivity over Zn2+ because of the high availability of Zn2+ in the natural environment. In this paper, bisquinoline-based fluorescent sensors with a 2-aminoethanol backbone were investigated. The weak coordination ability of quinoline compared to well-studied pyridine is suitable for Cd2+ selectivity rather than Zn2+. In the presence of 3 equiv. of metal ions, TriMeO-N,O-BQMAE (N,O-bis(5,6,7-trimethoxy-2-quinolylmethyl)-2-methylaminoethanol (3)), as well as its N,N-isomer TriMeO-N,N-BQMAE (N,N-bis(5,6,7-trimethoxy-2-quinolylmethyl)-2-methoxyethylamine (6)), exhibits Cd2+-selective fluorescence enhancement over Zn2+ in DMF-HEPES buffer (1:1, 50 mM HEPES, 0.1 M KCl, pH = 7.5) (IZn/ICd = 26–34%), which has similar selectivity in comparison to the corresponding ethylenediamine derivative TriMeOBQDMEN (N,N’-bis(5,6,7-trimethoxy-2-quinolylmethyl)-N,N’-dimethylethylenediamine) under the same experimental condition (IZn/ICd = 24%). The fluorescence mechanisms of N,O- and N,N-isomers of BQMAE are quite different, judging from the fluorescence lifetimes of their metal complexes. The Cd2+ complex with TriMeO-N,O-BQMAE (3) exhibits a long fluorescence lifetime similar to that of TriMeOBQDMEN via intramolecular excimer emission, whereas the Cd2+ complex with TriMeO-N,N-BQMAE (6) exhibits a short lifetime from monomer emission., 11 Jan. 2024, 29, 2, 369, 369, Scientific journal, 10.3390/molecules29020369

    1.不斉窒素原子と不斉酸素原子を有する銅錯体の合成と光学分割の試み 金属に配位することにより不斉窒素原子と不斉酸素原子を発生するアキラルな配位子およびさらに不斉炭素原子を導入したキラルな配位子を用いて、銅、パラジウム、白金などの金属錯体を調製した。得られた錯体のX線結晶構造解析により、金属に配位した不斉窒素原子の配置および不斉炭素原子に連動した不斉酸素原子の発現が確認された。現在引き続きHPLCおよびキラルなカウンターアニオンを用いた再結晶による光学分割を検討中である。
    2.イソキノリンの特性を利用した亜鉛およびカドミウムイオンの蛍光識別 4つのイソキノリン部位を有する化合物である3-isoTQLNは、同族元素である亜鉛イオンおよびカドミウムイオン存在下で異なる波長における蛍光増大を示した。他の金属イオン存在下では蛍光応答は見られなかった。対応するキノリン誘導体(TQLN)およびイソキノリン異性体(1-isoTQLN)との比較から、この興味あるデュアルセンシング機構のメカニズムの詳細を明らかにした。
    3.(イソ)キノリンの特性を活用したビスオキソ架橋マンガン二核錯体の構造と酸化還元特性 ピリジン、キノリン、イソキノリン、キノキサリン部位を有する15種類の配位子を用いてダイアモンドコア構造を有するビスオキソ架橋マンガン二核錯体を調製した。キラルなシクロヘキサンジアミン骨格を有する配位子ではホモダイマーのみが得られた。それらの酸化還元電位とX線結晶構造解析から得られた金属中心の配位環境および配位子の構造的特性との関係を詳細に議論した。従来広く用いられているピリジンとの比較により(イソ)キノリン配位子の特性を明らかにした。, kaken
  • Grant-in-Aid for Scientific Research (B), 01 Apr. 2016 - 31 Mar. 2020, 16H05656, Oversea's Research on Archaeological Proteins as Molecular Markers of Ancient Culture and Civilization, Nakazawa Takashi, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Nara Women's University, 15730000, 12100000, 3630000, The purpose of our project is to apply "protein archaeology" to proteins preserved in Paleolithic and Neolithic animal remains from archaeological sites in the area of western Asia and central Europe. For Neolithic animal bones, we identified animal species of bones and teeth by mass spectrometric amino acid sequencing of collagen. The animal species thus identified include cattle, deer, sheep, and goat. For the majority of Paleolithic animal teeth, however, there still remained ambiguity in the identity of species resulting from mass spectrometry because collagen was degraded so seriously that the quantity and number of peptide fragments obtained for the analysis were insufficiently small. To clarify the effect of such a long-term aging, we also investigated the degradation process of amino acids in proteins. One of the most interesting findings in our study is that the stability of triple-helical structure is particularly important for the longevity of type I collagen., kaken

By changing the linker length, nature and number of coordination atoms in the linker and substituents of the quinoline ring, as well as the replacement of quinoline with isoquinoline, we could improve the selectivity toward specific metal ions and anions. In our probe design, metal binding site and fluorescent moiety are combined in a molecule. The synthesized heavy metal targeted fluorescent probe was evaluated extra- and intracellular conditions.
    By careful design of the ligand scaffolds, several fluorescent probes that exhibit specific response for zinc, cadmium, and mercury have been developed. The present results provide an important roadmap for designing future fluorescent probes., kaken
  • Grant-in-Aid for Scientific Research (C), 2008 - 2010, 20550065, Chirality Control of "Asymmetric Oxygen Atom" generated by Metal Ion Coordination, MIKATA Yuji, Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Nara Women's University, 4290000, 3300000, 990000, In this project, by careful design of the ligand and the selection of metal ion, we are going to clarify the possibility of the chirality generated on the metal-coordinated oxygen atom and its limitation, proposing the brand-new concept of "Asymmetric Oxygen". By appropriate design of the ligand, we are going to isolate and characterize the enantiomer based on exclusively "Asymmetric Oxygen". At the same time, we are going to analyze the physical properties of the compound in order to find possible application of "Asymmetric Oxygen".
    At first, we examined the possible factors that are necessary to coordinate the achiral oxygen atom to the metal center. On the basis of this study, we constructed the system in which asymmetric oxygen and asymmetric nitrogen are generated simultaneously in the complex, and found that the configuration of the asymmetric nitrogen controls the configuration of the asymmetric oxygen. Furthermore, we succeeded in generating asymmetric oxygen atom using a ligand with an asymmetric carbon atom in various positions. X-ray crystallography of copper complexes revealed that the configuration of asymmetric oxygen controls the configuration of asymmetric oxygen atom. The chiral complexes exhibited characteristic Cotton effect in the d-d transition region of copper atom. But metal-coordinated nitrogen atoms have been demonstrated as an asymmetric nitrogen. Similarly, it is possible that the oxygen atom of a unsymmetrical ether can become a chiral center when coordinated to a metal ion. In this case also, the chirality is extremely unstable due to the rapid inversion of the remaining oxygen lone pair. Preventing lone pair inversion by metal ion coordination is a potential route to compounds exhibiting a chiral oxygen atom. Strong coordination of the metal ion to the ether oxygen atom may lead to the necessary differentiation of the two oxygen lone pairs. Careful ligand design is needed in any effort to isolate a compound with a metal-bound asymmetric oxygen atom. High Intense luminescent complexes with various emission colors were obtained rapidly with high yields. The photochemical properties of these luminescent complexes were investigated. Semiconductor HPA precise microwave chemical reactor has been newly developed for environment friendly reaction processes. With this equipment, microwave synthesis of metal complexes and nano gold particles has been efficiently carried out.

  • 若手研究(B), 2003 - 2004, 15750144, C-グリコシド結合を介した糖質への金属イオンの集積化と機能発現, 三方 裕司, 日本学術振興会, 科学研究費助成事業, 奈良女子大学, 3500000, 3500000, 本研究では、一般的な糖含有金属配位子の合成法の開発と、それを基盤とした糖質への金属イオンの集積化とその機能発現を行っている。生体にとって欠かせない糖質と金属イオンとの組み合わせによって、新しい物質領域を開拓することを目的としており、新しい合成手法と新機能物質の創製を目指す。本研究計画では,様々な糖を用いた一般的なC-グリコシド配位子の開発を行い,糖質の近傍に加水分解を受けにくい形で金属イオンを集積させることにより,水溶液中での不斉水素化触媒へ応用するなど,新しい物性の開拓と分子変換プロセスの開発を目指している。
    本研究により,C-グリコシド結合を有するジアミン配位子の合成法が確立された。概略を以下に示す。アセトブロモグルコースをジメチルホルムアミドを溶媒としてソジオマロノニトリルと反応させることにより,C-グリコシド結合を有するジシアノ体を合成した。次にこのニトリル部位の酸化白金を用いた接触水素化によりジアミン体を得た。続いて糖の水酸基の保護基として用いているアセチル基の脱保護をナトリウムメトキシドを用いて行ったところ,アミン部位へのアセチル転位が起こったが,脱保護を塩酸加水分解を用いて行うことにより,目的物である糖連結ジアミン体が高い収率で得られることがわかった。同様の合成ルートにより,ガラクトース連結ジアミン体の合成にも成功した。また,O-グリコシド結合を有する糖-フラーレン結合体および糖ジアミン-パラジウム錯体の生理活性,さらに糖ジピコリルアミン-銅錯体におけるアノマー酸素の配位による不斉酸素原子の発現について新しい知見を得た。, kaken
  • 萌芽的研究, 1999 - 2000, 11877373, 光線力学療法用ハイブリッド配糖ポルフィリンのデザイン合成, 矢野 重信; 三方 裕司, 日本学術振興会, 科学研究費助成事業, 奈良女子大学, 2100000, 2100000, 1.近年、ポルフィリン誘導体が光線力学的療法(PDT)に用いる光増感剤として注目されている。本研究では優れた光増感剤の開発を目指して生体的適合性に優れた糖分子を有する新規水溶性ポルフィリン誘導体のデザイン合成を行った。
    2.糖連結ベンズアルデヒド、ピロールと酢酸亜鉛を原料としてLindsey法を用いて糖の水酸基が保護されたZn-AcSugar TPPを調整し、それらを脱保護することにより、糖連結水溶性ポルフィリンを得た。Free-base AcSugar TPPは塩酸処理することによって調整した。また糖の水酸基が保護されたAcSugar TPPをp-toluenesulfonylhydrazideを用いて還元することにより、糖連結水溶性クロリンやバクテリオクロリンを調整し、それらを脱保護することにより、糖連結水溶性クロリン、バクテリオクロリンを得た。
    3.糖連結ポルフィリン、糖連結クロリン、糖連結バクテリオクロリンの合成に成功した。また、糖を連結させたことでバクテリオクロリンが純粋に得られ、バクテリオクロリン合成において初めての例となった。これらのクロリンは、UV-VISスペクトルにおいて長波長領域(650nm)、またバクテリオクロリンはさらに長波長領域(740nm)に大きな吸光係数のピークを持っており組織透過性の向上が期待される。すべての化合物は一重項酸素を発生し、Tetraphenyl porphine Tetrasulfonic Acid(TPPS)の約3倍の量子収率を示す化合物もあった。これらの培養がん細胞(HeLa細胞)に対する光毒性評価の結果、幾つかの化合物は従来にない優れた光毒性を示した。以上、本研究で得られた結果は、糖質による生体適合性に優れた光増感剤の設計の可能性を示す興味深い知見である。, kaken

A set of 1,3-propanediamine derivatives connected to carbohydrates has been prepared. D-Glucose, D-mannose, D-galactose, D-xylose, D-ribose, and D-maltose are utilized as sugar molecules in this work. The diamine moiety was connected to the Cl carbon of the glycopyranose ring via an O-glycoside bond. All the anomeric configurations and sugar puckering conformations, except in the D-maltose derivative, were determined by X-ray crystallography of the diazido or dibromo precursors. This method has been used to synthesize a library of sugar-pendant diamines including an OH-protected derivative, and an N,N'-diisopropyl-substituted derivative. Acidty constants of these sugar-pendant diamines have been examined, as have the stability constants of these ligands with the divalent metal ions in solution. The effective antitumor cisplatin type complexes of these sugar-pendant dimaines were developed. Sugar-dependent photocytotoxic property of tetra- and octa-glycoconjugated tetraphenylporphyrin and fullarene derivatives has been examined. Effective phototoxic property of newly prepared these glycoconjugated compounds was observed.   • 特定領域研究(A), 1999 - 2000, 11167253, ハイブリッド配糖体を機能素子とする分子シンクロシステムの光線力学療法への新展開, 矢野 重信; 覚知 豊次; 三方 裕司, 日本学術振興会, 科学研究費助成事業, 奈良女子大学, 3600000, 3600000, 1.本研究では光線力学的療法(PDT)に用いる優れた光増感剤の開発を目指して生体的適合性に優れた糖分子を有するFree-baseまたは亜鉛含有の新規水溶性ポルフィリン誘導体の合成を行った。
    2.糖連結ベンズアルデヒド、ピロールと酢酸亜鉛を原料としてZinc acetyl sugar tetraphenyl porphyrin(Zn-AcSugar TPP)を調整し、それらを脱保護することにより、糖連結水溶性ポルフィリンを得た。Free-base AcSugar TPPは塩酸処理することによって調整した。また糖の水酸基が保護されたAcSugar TPPを還元することにより、糖連結水溶性クロリンやバクテリオクロリンを調整し、それらを脱保護することにより、糖連結水溶性クロリン、バクテリオクロリンを得た。これらの光増感剤の一重項酸素発生能ならびに培養がん細胞(HeLa細胞)に対する光毒性評価を行った。
    3.糖連結ポルフィリン、糖連結クロリン、糖連結バクテリオクロリンの計80種類の化合物を合成した。また、糖を連結させたことでバクテリオクロリンが純粋に得られ、バクテリオクロリン合成において初めての例となった。特にバクテリオクロリンは長波長領域(740nm)に大きな吸光係数のピークを持っており組織透過性の向上が期待される。すべての化合物は一重項酸素を発生し。これらの培養がん細胞(HeLa細胞)に対する光毒性評価の結果、暗所では毒性を示さなかった。クロリン誘導体では糖水酸基を脱保護した水溶性化合物で飛躍的に活性が向上することがわかった。メタ置換亜鉛含有水溶性クロリン誘導体もfree-base体の同じく飛躍的に活性が向上することがわかった。また、メタ置換バクテリオクロリンの一部(m-TGlcPBCやm-TXylPBC)は700nm以下の光をカットしても優れた光毒性を示した。, kaken
  • 特定領域研究(A), 1999 - 1999, 11121221, 生理活性ハイブリッド配糖錯体の分子設計とバイオメカニズムの解明, 矢野 重信; 木下 勇; 三方 裕司, 日本学術振興会, 科学研究費助成事業, 奈良女子大学, 1300000, 1300000, 1.シスプラチンは極めて有効な抗腫瘍性薬剤として広く臨床に使われているが、難溶性、副作用の問題も多い。本研究では1,3-プロパンジアミンにO-グリコシド結合で糖と連結あさせた新しいタイプの配位子を開発した。それらを用いた糖連結シスプラチン型Pt(II)錯体を合成し、それらの制がん効果について検討した。
    2.D-glucoseのパーアセチル体と1,3-dibromo-2-propanolを原料として、5段階で合成を進め、新規配位子1,3-diamino-2-propyl b-D-glucopyranoside(2-β-D-Glc-pn)を合成した。同様の方法で1,3-diamino-2-propyl α-D-mannnopyranoside(2-α-D-Man-pn),1,3-diamino-2-propyl α(orβ)-D-galactopyranoside(2-α(orβ)-D-Gal-pn)を合成した。目的化合物の生成をNMR・MASSスペクトルの測定から確認した。糖質が連結したジアミンをK_2PtCl_4の水溶液と反応させ、[PtCl_2(2-β-D-Glc-pn)],[PtCl_2(2-α-D-Man-pn)],[PtCl_2(2-α-D-Gal-pn)],[PtCl_2(2-β-D-Gal-pn)]ならびに[PtCl_2(2-β-D-Mal-pn)]の合成を行った。[PtCl_2(2-β-D-Glc-pn)]のX線結晶構造解析を行い、他の錯体については元素分析、NMR、MASS測定によりその構造を明らかにした。Pt(II)錯体のマウス移植腫瘍(P388 cell)に対する制がん効果の検定を行った
    3.Glucoseを用いた配位子は、糖ユニットのピラノース環がβ-^4C_1アノマーのみ、mannoseはα-アノマーのみが単離され、galactoseはα-,β-アノマーの両方を得た。[PtCl_2(2-β-D-Glc-pn)]のX線結晶構造解析から、錯体は、配位子1分子、塩素2原子が白金1原子に配位した4配位平面型構造である。また、NMR測定から、溶液内での構造と結晶構造がよく対応していることが明らかとなった。各錯体のマウス移植腫瘍(P388 cell)に対するヒトがん細胞パネルによる制がん効果の検定の結果、[PtCl_2(2-α-D-Gal-pn)]がかなり大きな活性(T/C=194%)を示すことが明らかとなり、今後の発展が期待される。, kaken

Zinc(II) complexes containing N-glycosides derived from D-glucosamine (D-GlcN) and ethylenediamine (en), [Zn(D-GlcN-en)ィイD22ィエD2]XィイD22ィエD2(1a.X = Cl, 1b.XィイD22ィエD2 = SOィイD24ィエD2ィイD12-ィエD1), were prepared and characterized by elemental analysis and nuclear magnetic resonance spectroscopy involving X-ray crystallography, where D-GlcN-en = 1-{(2-aminoethyl)amino}-2-amino-1, 2-dideoxy-D-glucose. The complex cation of 1a has a nearly C2 symmetry and the central nickel atom is octahedrally coordinated by two tridentate N-glycoside ligands from D-glucosamine and ethylenediamine in a meridional mode. The result of n.m.r. measurements of 1a confirmed the similar structure of complex cation orevealed by X-ray crystallography.   • 萌芽的研究, 1998 - 1998, 10875180, 多核金属酵素を規範とする新規金属クラスター触媒による糖質の異性化, 矢野 重信; 三方 裕司, 日本学術振興会, 科学研究費助成事業, 奈良女子大学, 1. 本研究は新規ハイブリッド金属クラスターを設計し、生体の基幹物質である糖質をアルドースからケトースへの異性化を促進する新反応場の構築と作用メカニズムの解明を行い、さらには将来の新しい有機化学工業への展開を目的としている。
    2. trenと糖質(D-mannose,L-rhamnose)の脱水縮合によってN-グリコシド配位子(aldose_3-tren)を合成し、これにMnX_2(X=Cl,Br,NO_3,1/2SO_4)を作用させることにより錯体を合成した。単核錯体、橙色三核錯体及び紫色三核錯体が得られた。錯体は元素分析、UV-Vis、CD、磁化率、ESR、X線結晶構造解析によりキャラクタリゼーションを行った。
    3. X線結晶構造解析から単核錯体のMn^イオンはaldose_3-tren配位子によるC_3対称を持つob型構造のN_4O_3型7配位面冠八面体構造の糖質が互いに分子内水素結合で集合した閉じたかご型構造ob型構造ではをとっていることが判明した。橙色三核錯体はMn^Mn^Mn^の混合原子価イオンがほぼ直線上に並んだ三枝構造を有し、2分子の単核錯体が中心のMn^イオンを糖質でアルコキソ架橋している構造である。中心マンガンは3価には珍しい四角錐型5配位構造である。トリエチルアミン存在下で単核錯体から三核錯体が生成することより、2分子の単核錯体の糖質部分がMn^イオンを捕捉することによって三核錯体が生成するものと考えられる。紫色三核錯体の架橋糖質がアマドリ転移によりβ-フラノースに異性化した構造である。架橋糖質のみが異性化していること、アルコキソ架橋Mn三核構造が保持されていることから異性化は金属二核中心上で進行すると推定している。糖質が架橋した錯体の構造はキシロースイソメラーゼの推定メカニズムの構造に類似しており、酵素モデルとしてその反応機構を明らかにすることは大変興味深い。, kaken
  • 特定領域研究(A), 1998 - 1998, 10134223, 抗腫瘍性ハイブリッド配糖錯体の分子設計とバイオメカニズムの解明, 矢野 重信; 三方 裕司, 日本学術振興会, 科学研究費助成事業, 奈良女子大学, 1700000, 1700000, 1. 本研究では、光増感剤に高い細胞親和性を持たせ光線力学的療法(PhotodynamicTherapy,PDT)に応用することを目的として、分子中に糖分子を含む水溶性ポルフィリン・クロリンの合成とその細胞毒性の評価を行った。ポルフィリン骨格にはテトラフェニルポルフィリンを選択した。また、連結する糖分子としてN-アセチルグルコサミン、ガラクトースおよびグルコースを選択し、ポルフィリン・クロリン1分子あたり4分子あるいは8分子の糖を結合した以下のような光増感剤の合成を行い、その光照射下での殺細胞効果について検討した。
    2. 目的物の合成は、水酸基を保護した糖連結ベンズアルデヒドを原料にLindseyらの方法によってポルフィリンの合成を行い、各化合物の脱保護を行って、糖連結水溶性ポルフィリンを得た。また、各ポルフィリンを還元して糖連結水溶性クロリンへと導いた。
    3. 化合物のHeLa細胞に対する光毒性の評価を行った。薬剤とHeLa細胞を2時間接触させた後洗浄し、8分間光照射を行った後24時間培養した後の生存細胞数を調べ、光照射のみの時の生存細胞数を100%として化合物の光毒性を見積もった。水酸基を保護されたグルコースを連結した化合物(において、特に顕著な光毒性が観測された。興味深いことに、グルコースを結合した化合物では糖の水酸基の保護によって毒性が高まるのに対し、他の糖を有する誘導体では水酸基の保護の効果はほとんど観測されなかった。8つのグルコース単位を有し、非常に水溶性の高い誘導体はほとんど光毒性を示さなかった。以上のことより光増感剤に導入する糖の種類・水酸基の保護基の有無によって、細胞への取り込み量が変化し、光毒性に大きな影響を与えていることがわかった。現在、糖連結クロリンの光毒性について検討を行っている。, kaken

    2. (1)ルテニウム二核錯体中の架橋酢酸と安息香酸との新規交換反応:[Ru_2(dhpta)(μ-O_2CCH_3)_2]^+中の架橋酢酸と安息香酸の交換反応を検討した。反応は12時間後に完了した。この結果は修飾電極上で同様な交換反応が起こる可能性を強く示すものである。(2)金電極に連結用の安息香酸ユニットをもつ、新しいジスルフィド化合物の合成:クラスターの金電極に連結用の配位子としてHS-(CH_2)_n-O-C_6H_4-CO_2Hの開発を行い、さらにそのジスルフィドÅiCH_3O_2-C_6H_4-O-(CH_2)_6-S-)_2を合成した。
    (3) ジスルフィドの金電極への自己集積:ジスルフィドのエタノール溶液に金電極を浸し修飾電極を作成した。吸着量は還元的脱離法により見積もった。(4)架橋酢酸の置換反応による酸化還元活性なルテニウム二核錯体修飾電極の形成:メチルエステル化された修飾電極表面の安息香酸部を、NaOH溶液処理を行い安息香酸に変換した。この安息香酸部を[Ru_2(dhpta)(μ-O_2CCH_3)_2]^+の酢酸架橋部分との置換反応によりクラスター修飾電極を作成した。クラスターの連結をCV測定により確認した。
    3. 本研究で開発した方法は酢酸架橋をもつ様々な多核錯体を金電極表面上に容易に吸着する一般的な手段として、SAMと錯体自身の機能性を調べるのに極めて有用なシステムであり、今後の発展が期待される。, kaken
  • 奨励研究(A), 1997 - 1998, 09740492, 核酸認識能を有する抗腫瘍性白金錯体の開発と機能解析, 三方 裕司, 日本学術振興会, 科学研究費助成事業, 奈良女子大学, 2100000, 2100000, 従来より、ポルフィリンの誘導体がその高い腫瘍集積性から光線力学的療法に用いられる薬剤として注目を集めている。光線力学的療法(Photodynamic Therapy,PDT)は癌の治療法の一種で、予め患者に腫瘍集積性のある光増感剤を投与した後、患部にレーザー光を照射して癌細胞だけを選択的に壊死させるという方法であり、この十数年間に優れたレーザー機器が開発され、優れた癌の治療法として注目されている。有機合成化学的にも、現在、ポルフィリンに水溶性を持たせた化合物が盛んに合成され、そのPDTへの応用が模索されている。以上のことから我々は、水溶性とともに細胞に対する認識力を持たせることを期待し、ポルフィリンに8つのグルコースを導入した化合物を合成し、すでに合成法が報告されている4つのグルコース、N-アセチルグルコサミン、ガラクトース分子を含む化合物とともにその一重項酸素発生能および培養ガン細胞に対する光毒性について検討した。その結果、糖分子の種類および数によってその一重項酸素発生能はほとんど変化しないのに対し、HeLa cellに対する光毒性は大きく影響を受けることを見出した。特に3種の糖のうち水酸基がアセチル保護されたグルコースを4分子持つ誘導体は、他の糖分子を含む化合物に比べて10倍以上と特異的に強い光毒性を示した。今回の発見は、糖分子の種類・数・保護基の有無を適切に選択することにより、その光殺細胞効果を容易に制御することができることを示しています。
    またテトラフェニルクロリンに糖分子を導入した化合物も合成し、糖部位の保護基の有無によるHeLa cellに対する光毒性の効果は、ポルフィリン誘導体の場合とは異なり、適度な親水性を持つ誘導体に高い活性が発現することを見出した。, kaken

    2.ポルフィリン1分子あたり4分子あるいは8分子の糖を結合したポルフィリンの合成を行い、その光照射下での殺細胞効果について検討した。化合物は、NMRおよび紫外・可視吸収スペクトルにより確認した。次に化合物のHeLa細胞に対する光毒性の評価を行った。光照射時間に依存して明らかに殺細胞効果が大きくなることがわかった。次に、薬剤とHeLa細胞を2時間接触させた後洗浄し、8分間光照射を行った後24時間培養した後の生存細胞数を調べ、光照射のみの時の生存細胞数を100%として化合物の光毒性を見積もった。水酸基を保護されたグルコースを連結した化合物において、特に顕著な光毒性が観測された。興味深いことに、グルコースを結合した化合物では糖の水酸基の保護によって毒性が高まるのに対し、他の糖を有する誘導体では水酸基の保護の効果はほとんど観測されなかった。またこれらの化合物の光照射下での一重項酸素(^1O_2)発生能をジフェニルイソベンゾフラン(DPBF)の^1O_2による分解反応によって見積もったところこれらの化合物はヘマトポルフィリン(HP)と同程度の一重項酸素発生能を有し、さらに化合物間に顕著な光増感活性の差は認められなかった。
    3.以上より、化合物間の光毒性の差は主に細胞親和性に依存し、水酸基を保護したグルコースは特異的に細胞内に取り込まれやすいものと思われる。今後糖分子を連結したポルフィリンおよびより組織透過性の高い長波長領域の光を吸収するクロリンについても同様に光毒性の検討を行い、糖分子の種類、糖鎖の長さ、保護基の影響、化合物の水溶性および金属イオンの導入が光毒性に与える影響を調べる。さらに、動物試験を実施する予定である。, kaken
  • 重点領域研究, 1997 - 1997, 09237245, ノンヘム類似骨格を有するFeおよびRuクラスターによる電極表面の機能設計, 矢野 重信; 三方 裕司; 加藤 昌子; 棚瀬 知明, 日本学術振興会, 科学研究費助成事業, 奈良女子大学, 1800000, 1800000, 1.本研究では、新規ヘムエリトリン類似骨格を持つRuあるいはFe多核錯体を電極上に配向制御して整列させ、酸化還元反応における電子移動過程の制御ならびに反応メカニズムの解明を行い、さらに不斉酸化還元反応への応用への可能性を探索を目的としている。本年度は鉄(III)四核ならびRu(III)二核クラスターの合成ならびにクラスターの電極表面への連結のための新規化合物の開発を行った。
    2.1)鉄(III)四核錯体:アラニン錯体Na[Fe_4(dhpta)_2(O)(OH)(L-ala)_2]・6H_2Oを合成し、そのX線結晶構造解析を行った。μ-オキソ間に水素が存在していることがわかった。このことは、Robertらにより指摘された類似の構造を持つマンガン錯体について、Hoffmanらが提案している光合成系における水からの酸素発生モデルに関連して重要であると議論されている。本研究の鉄(III)四核錯体も類似の構造を持っていることから光合成系の酸素発生と関連して興味深い。2)Ru(III)二核クラスター:[Ru_2(μ-O)(μ-O_2CCH_3)_2(HBpz_3)_2]及び[Ru_2(μ-O)(μ-O_2CCH_3)_2(HBpz_3)_2]を合成しそれらのX線結晶構造を行った。3)[Ru_2(μ-O)(μ-O_2CCH_3)_2(HBpz_3)_2]の電気化学:CV測定により、μ-アルコキソおよびμ-フェノキソ架橋Ru二核錯体ではRu(II,III)混合原子価状態が、一方、HBpz_3を用いたμ-オキソ架橋ではRu(III,IV)混合原子価状態が安定に存在しうるということが判明した。また、プロトンの脱着によるクラスターの酸化還元挙動のチューニングの可能性を見いだした。これらはルテニウムクラスターを用いた酸化還元触媒の設計にとって有用な情報である。3)クラスターの金電極に連結用の配位子としてHS-(CH_2)_n-O-C_6H_4-CO_2Hを開発した。
    3.以上の様に、ノンヘム類似骨格を有する機能性金属クラスターによる電極表面の機能設計を目指す本研究の推進のための準備が整いいつつあり、今後の進展が期待される。, kaken

    (Mal)を用いてコバルト配糖錯体の合成を行い、得られた錯体の構造および酸素分子の着脱能を検討した。
    2.1)[{Co((D-Glc)_2-tren)}_2(μ-O_2)]X_3・5H_2O(X=Cl(1a),Br(1b))及び[{Co((Mal)_2-tren)}_2(μ-O_2)]Cl_3・6H_2O(2)を合成しそれらのX線結晶構造解析を行った。
    2)錯体1a,2を窒素下で紫外線光照射による酸素分子の着脱能を検討した。
    3.元素分析結果から1a・5H_2O、1b・5H_2Oならびに2・6H_2Oの組成であることが判明した。錯体カチオンは擬C_2対称の構造をとっており、IR、UV、CD、NMRから予想されたように2個のCo原子間に酸素分子が架橋する二核構造となっている。架橋酸素結合距離はO-O一重結合に対応するペルオキソ架橋であることが明らかとなった。
    錯体1a,2を窒素下で紫外光照射するとペルオキソ架橋二核錯体に特徴的な400nmの吸収が消失し、また照射後空気に触れると400nmの吸収がそれぞれ76%(1a)および26%(2)まで回復した。このことから錯体1a,2は紫外光照射によりCo(II)錯体とO2に変換し、また生成したCo(II)錯体は酸素を吸収して錯体1aおよび2を再生すると思われる。グルコース錯体1aはマルトース錯体2に比べて酸素架橋錯体の再生率が高いことから、糖の違いにより錯体の安定性に違いが生じたものと思われる。
    4.糖質の選択により酸素分子の接近を制御しうることを見いだした本研究は糖質を機能素子とする分子識別能を有する超構造錯体の設計にとって有用な情報を与えるものである。, kaken

    2.反応場としてのルテニウム二核錯体の混合原子価状態を解明しつつ、架橋配位子の化学修飾等により、適切な電子状態を形成する系を構築し電子移動過程の制御ならびに有機化合物の酸化還元反応における触媒活性能の発現を調べるために、ヘムエリトリン類似骨格をもつ一連の新規(μ-アルコキソ)二核ルテニウム(III)錯体,Na[Ru_2(dhpta)(O_2CR)_2](R=CH_3,C_6H_5,C_6H_4OH,C_6H_4,NH_2)の合成とキャラクタリゼーションを行った。X線構造解析の結果、立体化学が解明された。CVにより一電子還元体が安定に存在することが明らかとなった。磁化率の測定から各ルテニウムサイトの不対電子間に強い反強磁性的な相互作用があることが判明した。
    3.本年度はルテニウム錯体の基本的な性質を調べるに留まっているが、今後、電子構造の量子化学的検討、一電子還元体の単離を行い、さらには架橋配位子としてN,N′-2-ヒドロキシ-5-メチル-1,3-キシレンビス[N-(カルボキシメチル)グリシン]を用い、フェノキソ架橋された多核錯体を合成し、上述のアルコキソ錯体と比較検討したい。これらの情報を基盤に当初の目的を達成したい。, kaken
  • 一般研究(C), 1995 - 1995, 07805079, キラルな金属錯体反応場によるα-アミノ酸の光誘起不斉合成, 矢野 重信; 三方 裕司; 加藤 昌子, 日本学術振興会, 科学研究費助成事業, 奈良女子大学, 2000000, 2000000, 配位子の修飾により光増感剤としてキラルな環境場を制御したポリピリジンルテニウム錯体を用いて、アミノアルキルマロン酸あるいはその金属錯体を出発原料とするアミノ酸の新しい立体選択的光誘起反応を探索し、そのメカニズムを解明しつつ、高選択的な新しい反応系を構築することを目的として以下のような萌芽的な研究を行った。
    立体構造を制御するためのキラルな直鎖状多座配位子としてN,N´-ジエチル-1(R),2(R)-1,2-シクロヘキサンジアミン(N,N´-Et_2-chxn)およびアラニン(ala)の前駆体であるアミノメチルマロン酸(AMM)を配位子として含む[Co(AMM)_2(N,N´-Et_2-chxn)]^-の合成とキャラクタリゼーションを行った。ついで[Co(AMM)_2(N,N´-Et_2-chxn)]^-の酸性水溶液中での脱炭酸反応によりR_-アラニンとS_-アラニンの比が83:17という高度な選択性で不斉合成されることを明らかにした。また高度な立体選択性の要因を解明するために、〔Co(R-ala)_2(N,N´-Et_2-chxn)〕CIO_4を別途合成し、その構造をX線結晶構造解析により明らかにした。さらに新規な立体選択的光誘起反応系の構築にとって重要なキラルな修飾ポリピリジン_-ルテニウム錯体の開発の目的で、アルキル置換基を導入した修飾ポリピリジン_-ルテニウム(II)錯体を合成した。それらの光学分割を行い、得られた光学活性体の発光特性、光ラセミ化に対する安定性等の基本的な性質を調べた。今後本研究で得られたキラルな修飾ポリピリジン_-ルテニウム錯体を光増感剤として、また上記で合成した基質であるアミノアルキルマロン酸ならびにそれらのコバルト錯体の光誘起脱炭酸反応について励起ルテニウム錯体から基質への電子移動やエネルギー移動過程、逆電子移動過程、および両者の会合過程等における立体選択性を詳細に調べ、選択性の発現機構を明らかにしていく予定である。, kaken
  • 奨励研究(A), 1994 - 1994, 06854033, 二本鎖DNAとアミノ酸誘導体との塩基配列特異的相互作用, 三方 裕司, 日本学術振興会, 科学研究費助成事業, 奈良女子大学, 1100000, 1100000, 本年度は、まず最初にトリプトファン誘導体として、トリプトファンのインドール環をキノリン環に置換し、また側鎖をアミノメチル基に変換し、さらに側鎖の位置を変えた誘導体の設計・合成を行った。続いて、そのそれぞれについて、分光学的測定および融解曲線の考察から、それらの化合物が非常に弱くDNAと相互作用していることが明らかとなった。また、側鎖の位置によってDNAとの結合能力は大きく異なり、キノリンの8位に置換基を有するもののDNA結合能力が強いことも明かとなった。
    次に、これらの化合物について、DNaseIフットプリント実験を行った。その結果、DNAと相互作用の確認されたキノリン誘導体において、CT配列の一塩基上流での切断が、薬剤の濃度の増加に伴って大きくなった。つまりこのことは、トリプトファン誘導体がDNAのCT配列に特異的にインターカレーションし、その部分のDNA構造にねじれやゆがみを生じさせ、DNaseIによる切断を受けやすくしていると考えられる。このことから、アミノ酸のつであるトリプトファンにおいても塩基配列特異的な相互作用が期待でき、またタンパク質によるDNA認識においてもトリプトファンあるいはフェニルアラニン・チロシンのような芳香族アミノ酸残基がDNA認識に大きな役割を果たしている可能性のあることが示唆された。
    また現在、このトリプトファンのDNA塩基配列特異的な結合については、分子軌道計算によってその詳細が明かになりつつある。残念ながら現在のところは満足のいく結果が得られていないが、実験結果と計算結果の一致がみられるようになれば、計算機化学が今後のドラッグデザインにおいて大きな役割を果たすであろうことは疑う余地がない。さらに現在、この簡単な化合物を立体特異的に配置した分子の合成も検討中である。, kaken
